    Coverage and Energy Analysis of Mobile Sensor Nodes in Obstructed Noisy Indoor Environment: A Voronoi Approach

    The rapid deployment of wireless sensor network (WSN) poses the challenge of finding optimal locations for the network nodes, especially so in (i) unknown and (ii) obstacle-rich environments. This paper addresses this challenge with BISON (Bio-Inspired Self-Organizing Network), a variant of the Voronoi algorithm. In line with the scenario challenges, BISON nodes are restricted to (i) locally sensed as well as (ii) noisy information on the basis of which they move, avoid obstacles and connect with neighboring nodes. Performance is measured as (i) the percentage of area covered, (ii) the total distance traveled by the nodes, (iii) the cumulative energy consumption and (iv) the uniformity of nodes distribution. Obstacle constellations and noise levels are studied systematically and a collision-free recovery strategy for failing nodes is proposed. Results obtained from extensive simulations show the algorithm outperforming previously reported approaches in both, convergence speed, as well as deployment cost.Comment: 17 pages, 24 figures, 1 tabl

    Measurement of the spin temperature of optically cooled nuclei and GaAs hyperfine constants in GaAs/AlGaAs quantum dots

    Deep cooling of electron and nuclear spins is equivalent to achieving polarization degrees close to 100% and is a key requirement in solid state quantum information technologies. While polarization of individual nuclear spins in diamond and SiC reaches 99% and beyond, it has been limited to 60-65% for the nuclei in quantum dots. Theoretical models have attributed this limit to formation of coherent "dark" nuclear spin states but experimental verification is lacking, especially due to the poor accuracy of polarization degree measurements. Here we measure the nuclear polarization in GaAs/AlGaAs quantum dots with high accuracy using a new approach enabled by manipulation of the nuclear spin states with radiofrequency pulses. Polarizations up to 80% are observed - the highest reported so far for optical cooling in quantum dots. This value is still not limited by nuclear coherence effects. Instead we find that optically cooled nuclei are well described within a classical spin temperature framework. Our findings unlock a route for further progress towards quantum dot electron spin qubits where deep cooling of the mesoscopic nuclear spin ensemble is used to achieve long qubit coherence.     Recruitment of a SAP18-HDAC1 Complex into HIV-1 Virions and Its Requirement for Viral Replication

    HIV-1 integrase (IN) is a virally encoded protein required for integration of viral cDNA into host chromosomes. INI1/hSNF5 is a component of the SWI/SNF complex that interacts with HIV-1 IN, is selectively incorporated into HIV-1 (but not other retroviral) virions, and modulates multiple steps, including particle production and infectivity. To gain further insight into the role of INI1 in HIV-1 replication, we screened for INI1-interacting proteins using the yeast two-hybrid system. We found that SAP18 (Sin3a associated protein 18 kD), a component of the Sin3a-HDAC1 complex, directly binds to INI1 in yeast, in vitro and in vivo. Interestingly, we found that IN also binds to SAP18 in vitro and in vivo. SAP18 and components of a Sin3A-HDAC1 complex were specifically incorporated into HIV-1 (but not SIV and HTLV-1) virions in an HIV-1 IN–dependent manner. Using a fluorescence-based assay, we found that HIV-1 (but not SIV) virion preparations harbour significant deacetylase activity, indicating the specific recruitment of catalytically active HDAC into the virions. To determine the requirement of virion-associated HDAC1 to HIV-1 replication, an inactive, transdominant negative mutant of HDAC1 (HDAC1H141A) was utilized. Incorporation of HDAC1H141A decreased the virion-associated histone deacetylase activity. Furthermore, incorporation of HDAC1H141A decreased the infectivity of HIV-1 (but not SIV) virions. The block in infectivity due to virion-associated HDAC1H141A occurred specifically at the early reverse transcription stage, while entry of the virions was unaffected. RNA-interference mediated knock-down of HDAC1 in producer cells resulted in decreased virion-associated HDAC1 activity and a reduction in infectivity of these virions. These studies indicate that HIV-1 IN and INI1/hSNF5 bind SAP18 and selectively recruit components of Sin3a-HDAC1 complex into HIV-1 virions. Furthermore, HIV-1 virion-associated HDAC1 is required for efficient early post-entry events, indicating a novel role for HDAC1 during HIV-1 replication

    Molecular control of HIV-1 postintegration latency: implications for the development of new therapeutic strategies

    The persistence of HIV-1 latent reservoirs represents a major barrier to virus eradication in infected patients under HAART since interruption of the treatment inevitably leads to a rebound of plasma viremia. Latency establishes early after infection notably (but not only) in resting memory CD4+ T cells and involves numerous host and viral trans-acting proteins, as well as processes such as transcriptional interference, RNA silencing, epigenetic modifications and chromatin organization. In order to eliminate latent reservoirs, new strategies are envisaged and consist of reactivating HIV-1 transcription in latently-infected cells, while maintaining HAART in order to prevent de novo infection. The difficulty lies in the fact that a single residual latently-infected cell can in theory rekindle the infection. Here, we review our current understanding of the molecular mechanisms involved in the establishment and maintenance of HIV-1 latency and in the transcriptional reactivation from latency. We highlight the potential of new therapeutic strategies based on this understanding of latency. Combinations of various compounds used simultaneously allow for the targeting of transcriptional repression at multiple levels and can facilitate the escape from latency and the clearance of viral reservoirs. We describe the current advantages and limitations of immune T-cell activators, inducers of the NF-κB signaling pathway, and inhibitors of deacetylases and histone- and DNA- methyltransferases, used alone or in combinations. While a solution will not be achieved by tomorrow, the battle against HIV-1 latent reservoirs is well- underway

    Die Cochlea-Implantation hat sich in den letzten Jahren als Methode der Wahl in der Behandlung ertaubter Patienten etabliert. Hierdurch hat sich insbesondere für einige Kinder mit angeborener oder im frühen Kindesalter erworbener hochgradiger Schwerhörigkeit zunehmend die Möglichkeit ergeben, eine Regelschuleinrichtung zu besuchen. In der vorliegenden Studie sollte der schulische Werdegang aller im CI-Zentrum Bochum implantierten Kinder untersucht werden.47 in der Zeit zwischen 1994 - 2004 in Bochum mit einem Cochlea Implant des Models MED-EL Combi 40/ 40+ versorgte Kinder im Alter zwischen 6 - 17 Jahren wurden in die Untersuchungen eingeschlossen. 33 Kinder waren unilateral, 14 Kinder bilateral implantiert. Die Implantation erfolgte im Durchschnitt im Alter von 5,1J. (Spannweite: 1,6-11,8 J.). Die Eltern der Kinder wurden mittels eines 15 Punkte umfassenden Fragebogens zu Teilaspekten der schulischen Laufbahn ihrer Kinder befragt.5 von 14 bilateral versorgten und 4 von 33 unilateral versorgten Kindern besuchten zum Zeitpunkt der Befragung die Regelschule. Der Anteil der Kinder, der in einer Schwerhörigenschule unterrichtet wurde, lag mit 63% in der Gruppe der unilateral versorgten Kinder unwesentlich höher als in der Gruppe der bilateral versorgten Kinder mit 57%. 15% der im Alter zwischen 1,6-6,7 J. erstimplantierten Kinder besuchten eine Regelschule, wohingegen in der Gruppe der im Alter von 6,7-11,8 J. erstversorgten Kinder 21% in einer Regelschule unterrichtet wurden
